No, if the PCBs don't use -5v the jamma pin goes no where.

similar note

I have a similar type of question so I'll just append here...

If I run a jamma pcb on my big red 4 slot I need a -5V added in there correct?
If I just go lazy and get the JAMMA -> MVS adapter (also to save myself from having to keep unplugging my mvs all the time) does that take care of the -5V or do I still have the same issue?

MVS doesn't use -5v so you won't hurt anything.

but will i need to add in that line for the jamma.... it seemed you wouldn't with the adapter but then where does that source come from if you don't manually add one in...

Not all games use -5v so it's your call if you want to wire it in. I wired mine in just to have it there in case it's needed. It's best to hook it up in case you buy a board that needs it. Some boards that require -5v might have symptoms of other things like booting problems and graphical issues that you might not think are -5v related.
