Depends on the game.
A lot of the earlier titles especially loaded only once at the beginning of the game, and then not again. Also, if it's a lower meg count game, any loading that *is* there is minimal, and can be easily tolerated.
Where you run into problems is with the fighters. Every fighter that I played on the CD system had a load time of at least 10 to 12 seconds in-between rounds; higher meg games, such as Real Bout Special, had load times approaching 20 to 30 seconds.
It's not so bad if you're playing in 1-player mode, but it can get especially irritating in vs. modes.
Also, if you're the anal type, be aware that while the CD format offers some nice goodies, such as different (usually better) music, art galleries, that kind of thing, that there still usually is some missing animation. It can range from barely noticable, to quite a lot (in games like Real Bout Special, Real Bout 2, Last Blade, etc.).
Of course, if that doesn't bother you, or you've never seen the game in it's original format, then you've nothing to worry about!
