Well, let's see what's confirmed and make some standpoints on the topic.
NEW CHARACTERS
Tokugawa Yoshitora (guy with 1+4 swords, seems to have two stances as well)
Majikina Mina (cute gal with a bow and and a pet mascot following her)
YungFei (old chinese martial master)
KusareGedou (big gaki demon)
Rera (seems 'dark version' of Nakoruru)
Rasetsumaru (seems 'dark version' of Haohmaru)
Enja (seems 'dark version' of Kazuki - from Asura Zanmaden)
Suija (seems 'dark version' of Sogetsu - from Asura Zanmaden)
NOTE: all 'evil version' of character are totally different sprites, not a single frame reused.
NOTE2: As we know, Rurouni Kenshin artist Nobuhiro Watsuki did the character designs for three of the new characters. I bet those are Yoshitora, Mina and YungFei. We also know the in-game art of this game ROCKS! I wonder if Watsuki-sensei is responsible of the characters portraits as well...

(NOTE: returning character are basically the entire Amakusa Kourin cast, minus Amakusa and Zankuro. All sprites seems to have been re-coloured in a new and more detailed style - more similar to Zankuro Musouken. Characters do not seems to go 'red' as for the anger system).
BOSS CHARACTERS
Mid boss: (big barbarina-like guy, possibly also appearing as big blue monster-like guy)
Final boss: Kurokawachi Yumeji (androgynus guy almost hibiki-like - Ranmaru maybe?)
Secret boss (?): (Oda Nobunaga like guy)
Possible secret characters:
Amakusa Shirou Tokisada (sprite ready to be ripped from Sam3/4)
Minazuki Zankuro (sprite ready to be ripped from Sam3/4)
Earthquake (sprite rippable from SvC CHAOS)
Shiki (sprite rippable from SvC CHAOS)
Note on animations:
Animations of characters suffered of a severe cut rate from SSIII to SSIV. I wonder which level of animation will be implemented in this game.
