I've been working on reviving a friend's Punisher board and it plays great now, minus the fact that I get no sound. After doing some reading, I gather that this may be because it does not have an interface board to connect the q-sound board and the motherboard.

I found this page...
http://wiki.arcadeotaku.com/w/CPS1#CP_System_Dash
That details the pinouts for each of the connectors.

From what I can gather, I need to run the Jamma speaker + and - lines from the Q-Sound board CN9 to the mother board edge connector. Do I need to run 12v and ground lines between the two as well ? Any confirmation, help, or clarification would be greatly appreciated. Thanks.

I would connect the speaker +/- from the power amp on the Q board to the empty spaces on the A board's power amp section and then connect the ground and 12V planes between the boards. Wire to wire connectors would be a good idea just in case you need to pull the boards apart again.

Something like this....
